"To test if our purified TFIIH and pol II were transcriptionally active, we first combined both epitope-tagged multiprotein complexes with bacterially expressed recombinant (r) human TFIIB, rTBP, rTFIIE, rTFIIF, a supercoiled DNA template (pG5MLT, which contains five Gal4-binding sites preceding the adenovirus major late core promoter elements), and a linearized DNA template (pMLΔ53, which contains the adenovirus major late core promoter elements without the Gal4-binding sites).
