"To test if our purified TFIIH and pol II were transcriptionally active, we first combined both epitope-tagged multiprotein complexes with bacterially expressed recombinant (r) human TFIIB, rTBP, rTFIIE, rTFIIF, a supercoiled DNA template (pG5MLT, which contains five Gal4-binding sites preceding the adenovirus major late core promoter elements), and a linearized DNA template (pMLΔ53, which contains the adenovirus major late core promoter elements without the Gal4-binding sites). As shown in Fig.5 A, both immunoaffinity-purified pol II complexes were equally active in transcribing the DNA templates, in conjunction with f:TFIIH, purified from the ammonium sulfate fraction, and other GTFs (comparelane 1 with lane 10)."
