Rationale: The noradrenaline/norepinephrine transporter (NET) is an integral component of the plasma membranes in axons and axon terminals. In cultured SCG neurons, NET-immunoreactivity is localized to axons. ICC demonstrated co-localiation of NET with the synaptic vesicle marker synaptophysin, indicating a presynaptic localization (Fig.10). Pre-embedding immuno-EM using an antibody directed to an intracellular epitope revealed immunoreactivity for NET along the cytoplasmic side of the plasma membrane and within the cytoplasm on synaptic vesicles (Fig.11). In the study, a rabbit polyclonal NET antisera was raised against the peptide WERVAYGITPENEHHLVAQRDVR (amino acids 585–607, peptide N585). The specificity of the antibody was validated by immunoprecipitation, immunoblot and immunohistochemistry (Fig.1). No KO-control..
